US FDA White Paper on Key Considerations for the Development and Use of Digitally Derived Measures for Clinical Investigations
The US FDA has released a white paper on Key Considerations for the Development and Use of Digitally Derived Measures for Clinical Investigations.
The document focuses on outcomes that matter to patients to ensure the technology is fit-for-purpose. FDA emphasized that digitally derived endpoints must be validated in the actual study population, including relevant age groups, disease severity, and patient characteristics. Sponsors should define how missing data will be handled at every level of the trial, not just the patient level, to ensure endpoints are patient-centered and clinically meaningful. FDA stressed that a cleared device does not automatically validate a clinical endpoint.

EMA Releases Sponsor Guidance: New Safety Module on the Annual Safety Report
This EMA guidance introduces the new CTIS Safety Module for Annual Safety Reports (ASRs, formerly DSURs), which will become the mandatory submission route for new ASRs beginning 28 September 2026, enabling harmonized submission, assessment, and management of safety reports across EU/EEA clinical trials. It provides sponsors with detailed instructions on user roles, ASR creation and submission, management of Requests for Information (RFIs), transition from the legacy ASR process, and key system requirements and known issues within CTIS. This guidance is operational in nature and is intended to help sponsors prepare for the September 2026 CTIS Safety Module go-live.
The module introduces dedicated Safety roles (Sponsor Safety Admin and ASR Submitter) with role-based access controls.
The new CTIS Safety Module replaces the current ASR functionality, with a 2-month transition period ending 28 November 2026. Sponsors can submit either Clinical Trial-centric or Substance Group-centric ASRs, with SG-centric submissions expected to be used most frequently. After submission, ASRs undergo coordinated Member State assessment, and sponsors must respond to any RFIs within the defined timelines.
ASR documents and supporting materials submitted through the module are not publicly disclosed, although they are available to relevant EU Member State authorities.

Demonstrating Substantial Evidence of Effectiveness for Human Drug and Biological Products: Draft Guidance for Industry. Advances in our understanding of biological processes and the increasing availability of high-quality data have transformed the evidentiary landscape for drug development. Given these advances, the draft guidance discusses the many factors that can impact the strength of evidence of effectiveness for a drug and clarifies how sponsors can rely on one adequate and well-controlled clinical investigation with confirmatory evidence to satisfy the substantial evidence of effectiveness standard. When final, this guidance will replace the 1998 guidance titled "Providing Clinical Evidence of Effectiveness for Human Drug and Biological Products."
